Comparison of separation methods for immunomodulatory extracellular vesicles from helminths

چکیده
Abstract Helminths survive within their host by secreting immunomodulatory compounds, which hold therapeutic potential for inflammatory conditions. Helminth‐derived extracellular vesicles (EVs) are one such component proposed to possess activities. Due the recent discovery of helminth EVs, standardised protocols EV separation lacking. Excretory/secretory products porcine helminth, Ascaris suum , were used compare three methods: Size exclusion chromatography (SEC), ultracentrifugation (UC) and a combination two. Their performance was evaluated yield, sample purity ability EVs suppress lipopolysaccharide (LPS)‐induced inflammation in vitro. We found that all methods successfully separated with similar yield. Functional studies showed from reduced LPS‐induced levels tumour necrosis factor (TNF‐α) dose‐dependent manner. Overall, performance, however, UC+SEC presented slightly higher than either method alone.

Alternative Methods for Characterization of Extracellular Vesicles
Extracellular vesicles (ECVs) are nano-sized vesicles released by all cells in vitro as well as in vivo. Their role has been implicated mainly in cell-cell communication, but also in disease biomarkers and more recently in gene delivery. They represent a snapshot of the cell status at the moment of release and carry bioreactive macromolecules such as nucleic acids, proteins, and lipids. A major...
متن کاملMethods to isolate extracellular vesicles for diagnosis
Extracellular vesicles (EVs) are small membrane-bound bodies that are released into extracellular space by diverse cells, and are found in body fluids like blood, urine and saliva. EVs contain RNA, DNA and proteins, which can be biomarkers for diagnosis. EVs can be obtained by minimally-invasive biopsy, so they are useful in disease diagnosis. High yield and purity contribute to precise diagnos...
متن کاملCurrent methods for the isolation of extracellular vesicles.
Extracellular vesicles (EVs), including microvesicles and exosomes, are nano- to micron-sized vesicles, which may deliver bioactive cargos that include lipids, growth factors and their receptors, proteases, signaling molecules, as well as mRNA and non-coding RNA, released from the cell of origin, to target cells. EVs are released by all cell types and likely induced by mechanisms involved in on...
متن کاملThe Methods of Choice for Extracellular Vesicles (EVs) Characterization
In recent years, extracellular vesicles (EVs) have become a subject of intense study. These membrane-enclosed spherical structures are secreted by almost every cell type and are engaged in the transport of cellular content (cargo) from parental to target cells. The impact of EVs transfer has been observed in many vital cellular processes including cell-to-cell communication and immune response ...
متن کاملon the comparison of keyword and semantic-context methods of learning new vocabulary meaning
the rationale behind the present study is that particular learning strategies produce more effective results when applied together. the present study tried to investigate the efficiency of the semantic-context strategy alone with a technique called, keyword method. to clarify the point, the current study seeked to find answer to the following question: are the keyword and semantic-context metho...
